We (2 adults) want to spend 3 nights in the Trulli county and explore nearby towns via daily excursions. We have difficulty in deciding which to choose as a base (Matina Franca or Locorotondo). Which town do you recommend in terms of atmosphere, sights to see, nice food and accommodation, as well as easy access to bus/train stations?

You really can't go wrong with either one. Both have lovely white historic centers and a number of good restaurants, and train stations on the edge of town. However, Martina Franca is quite a bit bigger than Locorotondo, so perhaps would have the edge in having more options for dining, accommodations, etc. Also, Martina Franca has an enormous street market every Wednesday morning. No matter where you stay, try to get to Martina's market on Wednesday.
If you are going to be in Martina Franca in late July or early August, make sure to check out the Festival della Valle d'Itria. In addition to the Festival's classical concerts and opera performances (paid admission), there are many free concerts and events during that time.
My husband and I will also be staying outside of Martina Franca
early September. We are renting a car. We prefer to stay near lively towns with great food and drink, architecture, and people watching. Having a beach close by is a must! is Martina Franca a good choice for us?

Not sure what you mean by "a beach close by" -- is a 25 minute drive close enough? Because that's what it would be from Martina Franca to the nearby Adriatic beaches. It would be about the same distance to the Ionian coast at Taranto. But for great food and drink, lovely Baroque architecture, proximity to other places of interest (Alberobello, Locorotondo, Cisternino, Ostuni, Bari, Matera, Castellana Grotte), lots of bars on the piazzas for people watching, and a lively passeggiata in the evening, Martina Franca is a very good choice.

Mille Grazie, Puglia Vagabond. Any suggestions on what beaches are the best to visit from there?. Will the lidos be open in the first 2 weeks of September?
Sorry, I'm not much of a beach person so I don't have personal recommendations. I know that locals around here like Torre Canne and Savelletri beaches.
