When Eleonora Sophie Schleswig-Holstein-Sonderburg was born on 14 February 1603, in Sønderborg, Schleswig-Holstein, Prussia, Germany, her father, Hans II. der Jüngere von Schleswig-Holstein-Sonderburg, was 57 and her mother, Agnes Hedwig von Anhalt-Zerbst Prinzessin von Anhalt-Zerbst, was 29. She married Christian II von Anhalt-Bernburg Prinz von Anhalt-Bernburg on 27 February 1625, in Germany. They were the parents of at least 8 sons and 8 daughters. She died on 5 January 1675, in Ballenstedt, Anhalt, Germany, at the age of 71, and was buried in Bernburg, Saxony-Anhalt, Germany.
