Ursula St. Barbe was the daughter of Henry St. Barbe, of Ashington, Somerset, by his wife, Eleanor Lewknor, daughter of Edward Lewknor, of Trotten, Co. Sussex. She first married Sir Richard Worsley, by whom she had two sons, and following his death she was married in 1566 to Sir Francis Walsingham.

The following year, her two sons by Worsley, John and George, were killed along with others in an accidental gunpowder explosion at the Worsley estate on the Isle of Wight, Appuldurcombe. She had two daughters by her second marriage to Sir Francis Walsingham: Frances, who married Sir Philip Sidney; and Mary (1573-1580), who died in childhood.
