Doppio is a side Level 30+ quest NPC that will assign you a quest to defeat DIO. He can be found in the parking garage behind the parking lot where the Security Guards spawn.

Dialogue: Yes boss I uh...
1: Are you okay? You're talking to a frog. (continue) - ... (deny)
Dialogue: Oh uh, yeah! I need to kill "Deo". The boss says so.
2: I'll do it for you (continue) - Sorry, I can't. (deny)

If you talk to Doppio when you haven't killed DEO yet, then he will say:
"Bring me DEO's head."
