Charles initially performed stand-up comedy in the 1970s and eventually got hired to write for the sketch comedy show called Fridays. Here, he was able to work with Larry David and began his career in writing for television. He eventually wrote for the shows The Arsenio Hall Show and Seinfeld. Larry Charles eventually debuted as a director for the film Curb Your Enthusiasm.

Even if the show's episodes are mostly written by Jerry Seinfeld and David for the first five seasons, Charles was their second in command during this time. He became famous for contributing to the darker scenes and storylines of the show. His episodes also covered controversial topics like a hospital patient committing suicide, a psychotics stalker, and the Nazis. In an episode of the second season, he wrote: "The Bet" which concerned Elaine purchasing a handgun for protection. NBC did not film this because they thought that the use of the gun was too provocative. According to Charles, his written content on Seinfeld is heavily influenced by Abbot and Costello, Dragnet and Superman.
